If you are not sure there’s a second playthrough or not, do not ascend. If you have seen non-ascending Astarion and wanted to experience some unhealthy twisted relationship, ascend. This is what I’m doing in my current playthrough. So either choice can make for a great story, depending on what you're going for.

You don't have to do anything. They can't stop you from being good. I completed Astarion's romance as a goody-two-shoes paladin and had no problems.
